Background / About us:
The twin-arginine-translocation (Tat) protein translocation pathway has the remarkable ability to transport completely folded proteins across the bacterial cytoplasmic membrane. Due to this fact, this pathway is a very attractive alternative to the general secretion (Sec) pathway when a secretory production of heterologous proteins in bacterial hosts is envisaged. In the recent years, our group (i.e the “Bacterial Protein Secretion Group” at the IBG1) has significantly contributed to the understanding of the molecular mechanism of Tat-dependent protein transport in the Gram-negative model bacterium Escherichia coli. In addition, we have successfully used the Tat pathway for the secretion of various heterologous proteins by the industrially important Gram-positive bacterium Corynebacterium glutamicum.

Aims of the PhD-thesis:
We are seeking a highly motivated PhD student which will address the following topics in his/her PhD study:
- In depth analyses of substrate-translocase interactions and of functionally relevant interactions between Tat components in E. coli using genetic and molecular biological methods.
- Application of the generated knowledge for the optimization of Tat-dependent secretory production of heterologous proteins in C. glutamicum.

Your Profile:

  • Diploma or Master´s degree in Biology, Microbiology, Biochemistry, Biotechnology, or a related discipline.
  • Solid theoretical and practical knowledge of modern microbiology, molecular biology/genetics, and protein analysis/detection techniques.
  • Strong motivation, drive and enthusiasm, good communicative skills.
  • Ability to work in a proactive and independent way and to cooperate with other scientists in an interdisciplinary manner.
