Bachelor or Master Thesis: Regulation in Enzyme Cascade Reactions

Background:
The focus of the `Synthetic enzyme cascades´ group is the development of value-generating processes using enzyme cascades. We combine catalysts to multi-step biotransformations which do not occur conjunct in nature.

Project Description:
In such complex enzymatic cascades the occurrence of undesired side reactions and the balancing of the different reaction steps against each other are major challenges. The project aims to address both of this challenges:

  1. By means of enzyme entrapment in stimulus-sensitive microgels, the enzyme activity should be controllable by adjusting the swelling degree.
  2. A sensitive online-analytic method will provide direct reaction monitoring, to enable the balancing of the reaction steps.

Your tasks will include the immobilization of enzyme in stimulisensitive microgels, as well as the characterization of the loaded gels.

Furthermore, you are going to help in establishing an NMR-based online analytic for monitoring of the biocatalytic reaction.

The project can be started as soon as possible with the exact focus depending on the own interests and qualifications.

Your Qualifications:
You are a highly motivated student of biochemistry, applied chemistry, biotechnology, or a related field with (very) good grades and strong interest
in interdisciplinary research. Prior hands-on experience with biocatalysts and instrumental analytics (photometric assays, HPLC, and NMR) would be advantageous.
